BioNTTech's share price fell about 8% on Friday after the company terminated a Phase 2 study of autogene cevumeran, an mRNA cancer vaccine for colorectal cancer treatment.

A safety monitoring committee recommended early termination of the study due to futility concerns and differences in survival data between treatment groups.

This is the second trial failure of autogene cevumeran this year, after a bladder cancer study was terminated in March.

This news contrasts sharply with the recent positive momentum in the mRNA field, triggered by the results of Moderna and Merck's successful Phase 3 melanoma vaccine.

Despite setbacks, BioNTech maintains a solid balance sheet with 16.6 billion euros in current assets, while its pancreatic cancer research continues.

BioNTech shares fell about 8% on Friday after the biotech company and partner Genentech announced the termination of a Phase 2 clinical study of an mRNA cancer vaccine for colorectal cancer patients. Before the announcement, shares were trading around $104.

This clinical study evaluated autogene cevumeran as an adjunct to surgical intervention in patients with high-risk stage II or III colorectal cancer.

An independent data safety monitoring committee found differences in overall survival between the study treatment groups. The committee believes that continuing the test will be futile and there is little chance of changing the final result.

This study had previously reached the futility threshold in October 2025. However, at the time, the monitoring committee believed that the existing data lacked sufficient maturity to draw clear conclusions on the treatment effect.

This is the second setback suffered by autogene cevumeran. In March, BioNTech and Genentech terminated a bladder cancer study involving the same vaccine, citing evolving standards of treatment for the indication.

Comparison of the fate of the field of mRNA oncology

The timing of this announcement is particularly unfortunate. Just a week ago, BioNTech shares experienced their strongest trading day in six years after Moderna and Merck announced positive Phase 3 results for their mRNA melanoma vaccine used in conjunction with Keytruda.

These results pushed Moderna's share price up by about 177%, and had a rising tide effect across the mRNA sector. Friday's developments were a sharp reminder that clinical success for one cancer indication does not guarantee similar results for different tumor types.

The biological explanation behind this is related to tumor characteristics. Melanoma is an immunological "febrile" malignancy with a high mutation rate and usually responds to immunomodulatory therapy. In contrast, colorectal cancer is classified as a "cold" tumor that has historically been resistant to immunotherapy.

There are also significant differences in test methods. Moderna's study combined its vaccine with Keytruda, the established checkpoint inhibitor therapy. BioNTech evaluated autogene cevumeran as a monotherapy and set more stringent efficacy requirements.

BioNTech's future path

Professor Özlem Türeci, chief medical officer of BioNTech, described the results as disappointing, while emphasizing the scientific knowledge gained on the mechanisms of drug resistance in immunosuppressive tumors. She noted that these insights will inform the development of future mRNA cancer therapies.

Despite setbacks, BioNTech still has a solid financial foundation. The company disclosed it had 16.6 billion euros in cash reserves and marketable securities in the second quarter of 2026, despite a quarterly net loss of 820.8 million euros.

A clinical project is still ongoing. BioNTech's Phase 2 pancreatic cancer trial is evaluating autogene cevumeran in combination with checkpoint suppression therapy and chemotherapy and continues as planned.

Market participants are currently focusing on the ESMO conference scheduled for October 2026, when comparative data from BioNTech and Moderna projects may be released.

BioNTech also anticipates interim data readouts for its BNT113 study of head and neck cancer. Head and neck malignancies are sensitive to immunotherapy and may provide a more favorable environment for vaccine efficacy.

Roche, Genentech's parent company, fell about 1.2% on Friday due to the announcement.
